چکیده

Polarity-sensitive fluorescent probes like 8-anilino-1naphthalenesulphonate (ANS) and 1,1′-bis(4-anilino5-naphthalenesulphonic acid (bis-ANS) have been frequently used to detect equilibrium folding intermediates like the molten globule state, as the formation of the latter involves increase in hydrophobic exposure. The ability of these fluorescent probes to bind to the intermediate state thus provides a convenient method for detection of folding intermediates in a multiple-state folding transition. However, there is no convenient method available to quantitatively analyse the fluorescent changes detected as a function of denaturant concentration. Here we describe a new method for quantitative analysis that permits one to estimate free energy of folding and determine the relative population of the various states of protein in the solution.

d-PET-controlled “off-on” Polarity-sensitive Probes for Reporting Local Hydrophilicity within Lysosomes

Polarity-sensitive fluorescent probes are powerful chemical tools for studying biomolecular structures and activities both in vitro and in vivo. However, the lack of "off-on" polarity-sensing probes has limited the accurate monitoring of biological processes that involve an increase in local hydrophilicity. Here, we design and synthesize a series of "off-on" polarity-sensitive fluorescent probe...

The mode of α-synuclein binding to membranes depends on lipid composition and lipid to protein ratio.

Interactions of the presynaptic protein α-synuclein with membranes are involved in its physiological action as well as in the pathological misfolding and aggregation related to Parkinsons's disease. We studied the conformation and orientation of α-synuclein bound to model vesicular membranes using multiparametric response polarity-sensitive fluorescent probes together with CD and EPR measuremen...

Near infrared dyes as lifetime solvatochromic probes for micropolarity measurements of biological systems.

The polarity of biological mediums controls a host of physiological processes such as digestion, signaling, transportation, metabolism, and excretion. With the recent widespread use of near-infrared (NIR) fluorescent dyes for biological imaging of cells and living organisms, reporting medium polarity with these dyes would provide invaluable functional information in addition to conventional opt...

The use of coumarins as environmentally-sensitive fluorescent probes of heterogeneous inclusion systems.

Coumarins, as a family of molecules, exhibit a wide range of fluorescence emission properties. In many cases, this fluorescence is extremely sensitive to the local environment of the molecule, especially the local polarity and microviscosity. In addition, coumarins show a wide range of size, shape, and hydrophobicity. These properties make them especially useful as fluorescent probes of heterog...

Cholesterol: Revisiting its fluorescent journey on 200th anniversary of Chevruel's ``cholesterine''

The legacy of Micheal Chevruel’s discovery of “cholesterine” as a non-saponifiable lipid from gall stones has ignited the imagination and research of countless minds for over two centuries now. In this review, we have provided a brief chronicle of the early history of cholesterol research which paved the way to present day understanding of membrane biology. We have discussed the properties and ...
